Model-based analysis of the north activation, an p53 inhibition effect en metastasis

Both a gain of function of NICD1 and a loss of function of p53 have been shown to induce metastasis in mice. Due to the complexity of the two pathways Notch and p53, and their crosstalks, it is difficult to understand the mechanisms leading to metastasis. We constructed an influence network including the main components of the two pathways and other pathways known to induce metastasis to explain explain the phenotype of the double mutant.
We concentrated more particularly on the effect of NICD1 and p53 on the inducers of EMT. Using logical modelling, we were able to reconstruct the double mutant and other mutants in silico suggesting that the influence network is correctly recapitulating the mutant behaviour. Furthermore, we tried to associated experimental data to stable states, a way to validate the model network.
